THE ACHIEVEMENT IN A FIELD OF STUDY OF ORGANOFOSPHATES ACTION AND FORECAST OF DELAYED NEUROPHATY

Predictions of organophosphorus (OP) ester — induced delayed neurotoxicity (OPIDP) is one of the main tasks of modern medicine. The enzyme target of OPIDP is neurophathy esterase (NE), which is located in the nervoussystem. Activity similar to brain NE has olso been found in lymphocy of peripheral blood. The first biochemical lesion in the nervous system after OP treatment is significantly depressed NE in nervous tissue. More then 30 OP tests, with different structure in vivo and in vitro, were examined. Correlation has been found between inhibition NE in hen, guinea- pig and rat brains, as well as in lymphocytes from peripheral human blood. Ops such as fenitrothion, karbofos, malathion, dichlorvos etc., which have low inhibitory activity to NE (15-30 %), do not produce clinical disordes in hens. Electrophysiological investigation of the functional state of hens peripheral nervous system revealed that conduction velocity decrease along of nerves was an preclinical sign of OPIDP development. The inhibition of nonspecific enzymes activity (cholynesterase, carboxylesterase) in nervous tissu are not specific and cannot be used for prognostication of OPIDP. Search for medicines like reactivator of NE and OPIDP promotors was carried out in our ivestigation.
