THE METHOD FOR THE DETERMINATION OF THE RELATIONSHIP BETWEEN NITRIC OXIDE AND CYTOTOXIC INFLUENCE IN VITRO CONDITIONS

The method of study of nitric oxide role in the mechanism of chemical compounds toxic action to the cells in vitro conditions is developed.

The essence of the method consists in a comparison of an amount of death cells, incubated with researched chemical compound, under condition of use of cells, control and treated by a nitric oxide trap — the complex of diethyldithiocarbamate and iron (DTK-Fe2+). This complex is included in the cells, as a result of their successive incubation in mediums with sodium diethyldithiocarbamate and iron(II) sulfate. The developed method is approved on an example of the cytotoxic action of sodium nitroprusside and nitroglycerine, as well as N-nitrosodimethylamine on a model of rats' activated peritoneal macrophages. It was determined that the presence of the NO trap in macrophages structure did not have a significant effect on N-nitrosodimethylamine cytotoxic action, but almost completely eliminated the cytotoxic action of sodium nitroprusside and to the lesser extent nitroglycerine.
